Chicken IgY is a passive interference blocker that works by preventing anti-chicken antibodies from binding to the antibodies in the assay and causing a false negative or false-positive result. It must be added in excess concentration so that enough alternative binding sites are provided to remove weakly binding antibodies that can cause interference. Chicken IgY should be used in a sandwich or competitive ELISA or lateral flow assays that use a chicken antibody. Any human anti-chicken antibodies in the patient sample that could potentially interfere with the assay signal can bind to the chicken IgY, preventing non-specific binding that could interfere with assay results.
