等温扩增

等温扩增(包括HAD、MCA和LAMP)是一种在即时诊断(POC)中发挥着越来越重要作用的DNA扩增技术。 与qPCR和其他扩增技术相比,它具有快速、灵敏、低成本、方便等方面的优势,是在实验资源有限地区进行即时检测的理想选择。 等温扩增已被广泛应用于病原体检测(如沙门氏菌和疟疾),转基因作物污染,以及法医检测等。

以环介导等温扩增LAMP为例,它使用4到6个引物来识别目标DNA中的几个特定区域。 该反应由链置换聚合酶(如Bst DNA聚合酶)和两个特别设计的引物启动,形成环状结构,然后进行后续快速大量的DNA扩增。 LAMP反应对样本中的抑制物有较强耐受性能,能在短时间内产生大量的DNA,可以通过裸眼观察(如沉淀或颜色变化)。 具体来说,1 ~ 10个拷贝DNA可在30分钟内扩增到10^9 ~ 10^10个拷贝,9 to 1010 具有高灵敏度和特异性。
