等温扩增

等温扩增(包括HAD、MCA和LAMP)是一种在即时诊断(POC)中发挥着越来越重要作用的DNA扩增技术。 与qPCR和其他扩增技术相比，它具有快速、灵敏、低成本、方便等方面的优势，是在实验资源有限地区进行即时检测的理想选择。 等温扩增已被广泛应用于病原体检测（如沙门氏菌和疟疾），转基因作物污染，以及法医检测等。

以环介导等温扩增LAMP为例，它使用4到6个引物来识别目标DNA中的几个特定区域。 该反应由链置换聚合酶(如Bst DNA聚合酶)和两个特别设计的引物启动，形成环状结构，然后进行后续快速大量的DNA扩增。 LAMP反应对样本中的抑制物有较强耐受性能，能在短时间内产生大量的DNA，可以通过裸眼观察(如沉淀或颜色变化)。 具体来说，1 ~ 10个拷贝DNA可在30分钟内扩增到10^9 ~ 10^10个拷贝，9 to 1010 具有高灵敏度和特异性。

迈迪安还开发了特定样本类型的抗抑性LAMP预混液，设计用于从粗裂解液或富含抑制剂样品的直接扩增，具有更强的抑制物耐受性能。这些预混液可以用于液体体系，也可以通过冻干或风干开发常温稳定的检测用于室温运输和存储，延长保质期，并增加加样量提高样本灵敏度，是开发POCT快速检测的理想选择。
