Meridian’s Liquid Stable Flex RNA/DNA qPCR Mix (MDX988) is a high-concentration (5x), liquid one-step RT-qPCR master mix that delivers consistent, reliable results without the need for cold storage. Designed for flexibility, it can be stored and transported at ambient temperature for up to 10 months, helping assay developers reduce costs, simplify logistics and support more sustainable molecular testing workflows.

Supplied without primers and probes, MDX988 gives end users the freedom to add their own oligonucleotides at the point of use. This makes one master mix suitable for multiple targets, panels and development paths while avoiding the cold-chain burden of conventional frozen RT-qPCR reagents.

Liquid Stable Flex RNA/DNA qPCR Mix (MDX988) was stored at –20°C (control) or ambient temperature at 20°C for 10 months and then tested in triplex RT-qPCR assay with two template dilutions. These results indicate that compared to a frozen master mix, the Liquid Stable Flex RNA/DNA qPCR Mix stored at ambient temperature showed no significant loss in performance, as measured by fluorescence intensity and Ct values, demonstrating that Liquid Flex Ready RNA/DNA qPCR Mix is stable at ambient temperature for up to 10 months.

Performance of MDX988 was evaluated against Vazyme’s AccurSTART II U+ One Step RT-qPCR Super Premix in a triplex RT-qPCR assay using two input concentrations of mammalian total RNA (1.5 ng and 15 pg). MDX998 demonstrated earlier Ct values across the three targets, indicating greater sensitivity. In addition, MDX988 showed lower standard deviation between replicates, highlighting superior reproducibility and overall assay performance compared to the competitor formulation.

How should assays be prepared?
MDX988 is supplied without primers and probes. Add the appropriate primers, probes and template at the point of use according to the Product Handling Guide. Use sterile plasticware and nuclease-free reagents. If oligonucleotides need to be rehydrated, use nuclease-free sterile water and handle assembled reactions under sterile conditions, for example, using a laminar flow hood.
Yes. Once primers and probes are added, assembled reactions are bench-stable for up to 4 hours at ambient temperature. For long-term ambient storage of assays preassembled with primers and probes, use a ready-format product such as Liquid Stable Ready RNA/DNA qPCR Mix (MDX998).
Yes. Shipping and temporary storage for up to 10 months at 20 °C has no detrimental effect on the quality of Liquid Stable Flex RNA/DNA qPCR Mix when handled according to recommended guidelines. Transit and storage time should be considered when determining remaining self-life.
qPCR can amplify tiny amounts of DNA, so even small amounts of contamination can produce false positives. Contamination can arise from other samples, the laboratory environment or carryover amplification products from previous reactions. MDX988 includes dUTP for workflows designed to incorporate uracil into amplification products and can support UDG/UNG-compatible carryover-control strategies. 
Shipping and temporary storage for up to 10 months at ambient temperature, 20 °C, has no detrimental effect on product quality. For long-term storage, store at -20 °C and avoid repeated freeze/thaw cycles.
