Meridian’s Liquid Stable Ready RNA/DNA qPCR Mix is a one-step RT-qPCR master mix formulated for long-term ambient storage.

Designed for modern molecular workflows, MDX998 enables assays to be pre-assembled with primers and probes and stored at ambient temperature for up to 10 months without lyophilization, drying or rehydration. The mix maintains performance for up to 10 months at 20 °C, 1 month at 37 °C or 12 months at 4 °C, with or without primers and probes added.

Meridian’s Liquid Stable Ready RNA/DNA qPCR Mix is a high-concentration, ambient-stable liquid one-step RT-qPCR master mix designed for molecular assay development and manufacturing.

The formulation delivers consistent amplification performance throughout its shelf life and supports standard, fast cycling and multiplex RT-qPCR workflows. Assays assembled with primers and probes can be stored and shipped at ambient temperature without freezing, drying, lyophilization or rehydration.

Liquid Stable Ready RNA/DNA qPCR Mix (MDX998) was stored at –20°C (control) or ambient temperature at 20°C for 10 months and then tested in triplex RT-qPCR assay with two template dilutions. These results indicate that compared to a frozen master mix, the Liquid Stable Ready RNA/DNA qPCR Mix stored at ambient temperature showed no significant loss in performance, as measured by fluorescence intensity and Ct values, demonstrating that Liquid Stable Ready RNA/DNA qPCR Mix is stable at ambient temperature for up to 10 months.

Liquid Stable Ready RNA/DNA qPCR Mix, 5x (MDX998) was used in a 10-fold serial dilution of viral RNA target (10,000, 1,000, 100, 10 and 1 copy, respectively). The results illustrate that MDX998 offers exceptional sensitivity (down to 1 copy per reaction), reproducibility and qPCR performance.

How should the premixed assay be prepared?
Since the master mix and the premixed assays can be stored at ambient temperature for at least 10 months, it is critical to use sterile plasticware and reagents (i.e. tubes, oligonucleotides). If needed, we recommend rehydrating the required oligonucleotides in nuclease-free, sterile water. The recommended volume of primers and probes to be added to MDX998 is described in the Product Handling Guide. The assembled assays (Assay Ready RNA/DNA qPCR Mix, primers and probes) should be handled under sterile conditions, for example, using a laminar flow hood. The ready-to-use assays dispensed in tubes should be sealed with air-tight caps to prevent evaporation. We recommend storing them away from light to avoid bleaching the fluorophores that may be used. 
Yes. Assays assembled with MDX998, primers, and probes can be stored and shipped at ambient temperature when handled according to recommended guidelines. Because shipping time contributes to the overall validated shelf life, customers should consider transit and storage conditions when determining the remaining shelf life available to end users. For applications requiring the maximum available shelf life upon receipt, refrigerated or frozen shipping may be preferred.
As qPCR can amplify tiny amounts of DNA, preventing contamination is essential, as even small amounts of qPCR contamination can produce false positives. Cross-over qPCR contamination can include cross-contamination from other samples, contamination from elsewhere in the laboratory, and carryover qPCR contamination from amplification products. The latter causes most of the false positive results seen in qPCR. Preventing contamination can be difficult and so laboratory procedures must be in place to ensure DNA from one assay is not re-amplified in subsequent assays.
Shipping and temporary storage for up to 10 months at ambient temperatures (20°C) has no detrimental effects on the quality of Liquid Stable Ready RNA/DNA qPCR Mix. However, for long-term storage we do recommend storing at -20°C. Repeated freeze/thaw cycles should be avoided.
When stored and handled under the recommended conditions, full activity of the mix (without the primers and probes) is retained until the Expiry Date printed on the label.
