STI diagnostics are based on detecting either pathogen-derived targets or the host immune response to infection. This is achieved through two primary approaches: immunoassays, which detect antigens or antibodies, and molecular methods, which detect pathogen nucleic acids.

Immunoassay-based techniques enable the detection of pathogen-specific antigens or host antibodies (IgM, IgG), supporting screening, diagnosis, and disease monitoring across formats such as ELISA, CLIA, lateral flow, bead-based assays, and IHC.

Molecular methods, including nucleic acid amplification tests (NAATs) such as PCR and isothermal amplification, provide highly sensitive detection of pathogen genomic sequences and are often used alongside immunoassay-based approaches in modern diagnostic workflows.
