The glycerol-free mixes contain optimized excipients compatible with freezing and lyophilization, making them ideal for POC urine testing.

Urinary tract infections (UTIs) affect over 150 million people worldwide and are the second most common cause of infection in primary and secondary care and most women experience at least one episode of acute uncomplicated cystitis in their lifetime. UTIs are responsible for a major part of antibiotic prescriptions, accounting for up to 15% of antimicrobial use in the community, with antibiotic use been described as one of the main factors contributing to the emergence of antimicrobial resistance. With the stigma attached to sexually transmitted diseases (STDs), and with their rapid spread in the last few years, there is also a need for discrete, fast and accurate point-of-care (POC) tests as well.

The Lyo-Ready Direct DNA LAMP Urine and Lyo-Ready RNA/DNA LAMP Urine mixes combine even greater tolerance with fast, robust isothermal amplification to enable highly sensitive direct detection using crude urine samples. The mixes are uniquely designed to overcome much higher concentrations of inhibitors and so do not require a urine DNA or urine RNA purification step analyse a patient sample. Very fast reaction kinetics result in faster time to results and superior amplification sensitivity and specificity. In addition, the 4x concentrated master mixes are formulated with lyo-excipients which are optimized for freeze-drying to create ambient-temperature stable assays, making them ideal for POC urine testing.

FAQs: LAMP Urine Test

The main difference between loop mediated isothermal amplification and qPCR is that in loop mediated isothermal amplification, the amplification is achieved using a single constant temperature, while in the qPCR, amplification requires cycling between multiple temperatures on a thermocycler. Instead of melting DNA strands apart at high temperatures, loop mediated isothermal amplification uses polymerases with high strand displacement activity, like Bst DNA polymerases. As these polymerases do not need multiple temperatures, they can amplify a target in less than an hour, and in some cases in as little time as 10 minutes.

The advantages of an isothermal amplification urine test is that it is completely non-invasive, and it can be self-sampled in large quantities. Urine testing is used in the diagnosis and management of infectious diseases (including STDs) and urinary tract infections (UTIs).

The most common infections are urinary tract infections (UTIs), most commonly E. coli, P. mirabilis, Enterococcus, Staphylococcus and Klebsiella. This is followed by sexually transmitted diseases (STDs) such as syphilis, gonorrhoea, chlamydia, trichomoniasis, hepatitis B (HBV), herpes simplex virus (HSV), HIV and human papillomavirus (HPV). Several other infections can be diagnosed by urine DNA and urine RNA tests including community-acquired pneumonia (CAP), legionellosis, tuberculosis, congenital cytomegalovirus (CMV) infection, and arboviruses, such as Chikungunya virus, dengue virus and Zika virus. Parasites can also be diagnosed from urine testing for the detection of eggs from Schistosoma haematobium.

We have used up to 30% human urine with our mix.

It is stable for up to 2 years at ambient temperature if correctly stored in sealed pouches.

That will depend on the bacteria, for some bacteria you may not need to do any pre-treatment, however some may require heating to 95°C for 5 min and for some tougher bacteria, a Proteinase K step (Proteinase K 55°C for 15 min and then at 98°C for 5 min) or using a non-ionic detergent (such as Tween-20 to final concentration of 0.2% followed by incubation at room temperature for 10 min) may be necessary.

We do not see a performance difference between the two mixes, which is why we call the Lyo-Ready Direct RT-LAMP Urine mix – Lyo-Ready Direct RNA/DNA LAMP Urine mix, as it can be used for both.

Yes, the Lyo-Ready Direct DNA LAMP Urine and Lyo-Ready Direct RNA/DNA LAMP Urine can be used as a liquid mix or lyophilization and stored for up to 24 months without needing to change the reaction conditions and lyophilization/ambient temperature storage will not affect the sensitivity of the test.
