Optimized Lyo-Ready Master Mixes for Superior Diagnostic Assays
Meridian’s lyo-ready master mixes are the market-leading solution for molecular diagnostic assays requiring fast cycling, superior sensitivity, high inhibitor tolerance and ambient temperature stability.

Meridian master mixes combining the lasts advances in buffer chemistry and PCR enhancers, together with an optimized concentration of enzymes, dNTPs and MgCl2. and lyophilization compatible excipients. They come as standard as well as specimen-specific qPCR and RT-qPCR master mixes, developed for highly reproducible, accurate RNA and DNA target amplification, delivering excellent results in multiplex assays, with sensitivity down to a single copy.

Lyophilization (freeze-drying), is a process of removing the water content from a substance by sublimation after it has been frozen, while preserving its structure and properties. The reagents can be lyophilized to a bead or a cake format.
Lyophilization helps to extend the product’s shelf life, enhance the stability of the reagents at higher temperatures, and allows for convenient and accurate dispensing of the reagents. After lyophilization, the reagents are easily reconstituted with sample material.
We do not see a performance difference between the two mixes, which is why we call the Lyo-Ready Direct RT-qPCR master mixes – Lyo-Ready™ Direct RNA/DNA qPCR master mixes, as it can be used for both.
The Lyo-Ready™ qPCR and RT-qPCR master mixes have some inhibitor tolerance, but for amplification from crude lysates or inhibitor-rich samples we have the Lyo-Ready sample specific (blood, saliva, urine and stool) mixes.
Yes, reaction conditions are the same and they will produce the same Ct values, even with multiplex qPCR assays. This means that the liquid mixes can be used to create SOPs if you do not want to lyophilize and then later if lyophilization is required (for example to increase sensitivity), the SOP can be updated for lyophilization, it does not require completely new SOPs to be written.
Following lyophilization in the presence or absence of primers and probes, the mix is stable for a minimum of 24 months at ambient temperature (17 – 23 °C). Following rehydration, the assay reproducibility, sensitivity, and robustness will be the same as for a freshly made liquid mix, making the mixes ideal for point-of-care molecular diagnostic tests and microfluidic qPCR devices.
No, our lyo-excipients are proprietary, if you require a buffer that does not contain lyo-excipients, we have the Lyo-Ready™ qPCR Buffer w/o Excipients, 4x (MDX061), but you will need to supply your own excipients.
